چکیده

Introduction Microscopic polyangitiis is characterized by anti-neutrophil cytoplasmic antibodies (ANCA) and small vessel vasculitis. The diseases usually respond to cyclophosphamide (Cyc) but some patients are therapy resistant. Rituximab (RIT) is a chimeric antibody to CD20 causing lysis of B-lymphocytes and used for treatment of lymphomas. RIT has also been successfully used in patients with rheumatoid arthritis (RA), idiopathic thromobocytopaenic purpura, autoimmune haemolytic anaemia, cold agglutinin disease, systemic lupus erythematosus and vasculitides.

Place in therapy of rituximab in the treatment of granulomatosis with polyangiitis and microscopic polyangiitis

Granulomatosis with polyangiitis and microscopic polyangiitis are small vessel vasculitides characterized by circulating antineutrophil circulating antibodies. Standard treatment for active severe disease has consisted of cyclophosphamide with glucocorticoids with or without plasmapheresis, which achieves approximately 75% sustained remission, but carries significant adverse effects such as mal...

Current therapy of granulomatosis with polyangiitis and microscopic polyangiitis: the role of rituximab

Granulomatosis with polyangiitis and microscopic polyangiitis are anti-neutrophil cytoplasmic antibody-associated vasculitides (AAVs) that are prone to cycles of remission and relapse. The introduction of cytotoxic therapy has changed the prognosis for these diseases from typically fatal to manageable chronic illnesses with a relapsing course. Despite improvements in outcomes, recurrence of dis...

Treatment of ANCA-associated vasculitis: new therapies and a look at old entities.

Anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV) is a small-vessel vasculitis that primarily comprises 2 clinical syndromes: granulomatosis with polyangiitis and microscopic polyangiitis. Cyclophosphamide and glucocorticoids have traditionally been used for induction of remission. However, more recent studies have shown that rituximab is as effective as cyclophosphamide f...

Rituximab therapy in pemphigus and other autoantibody-mediated diseases

Rituximab, a monoclonal antibody targeting the B cell marker CD20, was initially approved in 1997 by the United States Food and Drug Administration (FDA) for the treatment of non-Hodgkin lymphoma. Since that time, rituximab has been FDA-approved for rheumatoid arthritis and vasculitides, such as granulomatosis with polyangiitis and microscopic polyangiitis. Additionally, rituximab has been used...
